Abstract Objective To investigate the direct effect of X-ray radiation on activity and apop tosis of cultured rat cardiomyocytes in vitro.Methods The study was performed using primary cell cultures of cardiomyocytes isolated from hearts of now-born rats.After being cultured for 4 days in vitro,cardiomyocytes were irradiated with single dose of 10Gy、20Gy、30Gy、40Gy of X-ray respectively.viability of cardiac muscle cells were assessed by MTT colorimetric assay,Myocardial of lactic acid dehydrogenase (LDH)was examined by blood biochemical autoanalyzer.Meanwhile,the Morphological characters of apoptosis were checked with laser-scanning confocal microscope by AO/PI staining and cardiac myocytes apoptosis rates were detected by flow cytometry.Results ①X-ray inhibited the growth of rat cardiac muscle cells in the dose-dependment or time-dependment manner in a certain range of dose.②the Morphological characters of apoptosis induced by X-ray (40Gy)were surveyed by laser-scanning confocal microscope in cultured rat cardiac cells.③X-ray induced apoptosis rate was detected being increased by flow cytometry.Conclusion X-ray has toxic effect,causing the damage of cardiac cells or apoptosis.
