    Objective To synthesize genes of the heavy chain variable region (VH) and light chain variable region (VL) of single-chain antibody of human cardiac myosin heavy chain for the development of myocardial imaging agents:single-chain antibody of human cardiac myosin heavy chain. Methods To extracted total RNA of the anti-HCMHC McAb hybridoma using TRIZOL reagent, synthesize the first-strand cDNA, using this first-strand cDNA as template, with specific primers, DNA polymerase and four single nucleotides, amplifiy the genes of the heavy chain variable region (VH) and light chain variable region (VL) by PCR. To identifiy products of each step and study relationship between RNA stability and storage temperature and optimize cycle selection temperature with MgCl2 concentration. Results The purity of the first-strand cDNA reached 95%, PCR products by agarose gel electrophoresis showed a single band with a bright swimming, molecular weight of VH and VL genes were 340bp and 320bp, which was consistent with literature reports. Conclusion Synthesized genes of VH and VL, laid the foundation for the development of myocardial imaging agents:single-chain antibody of human cardiac myosin heavy chain.

    Objective To get the X-ray spectrum under radiotherapy, diagnosis and protection fields. Methods The simulations were performed using different combinations of filters and tube Voltages with Geant4. All the relevant physical processes were included in the calculations:Compton scattering, photoelectric effect, Rayleigh scattering, bremsstrahlung and ionization. Results The X-ray spectrum and mean values of the spectrum under radiotherapy, diagnosis and protection fields. Conclusion The X-ray spectrum under radiation, diagnostic and protection fields can be acuately simulted with Geant4.

    Objective To explore the suppression function of healthy products mainly made from rana japonica oil of different dosage configuration compound prescription powder grown in the Changbai Mountain on oxidation resistance effect of the big mice which have been under the condition of overload and radiation. Methods The models of wister big mice under overload and radiation were established and intervened with the healthy products made from rana japonica oil and squalene. the ability of total oxidation resistance、superoxide dismutase、maleic dialdehyde and the hydroxyl free radical in the serum taken from ead group of big mice was examined. Results The total oxidation resistance ability's activeness of low dose rana japonica oil line is obviously higher than that of other 5 groups. The ability of the overload radiation group's maleic dialdehyde is obviously higher、the hydroxy free radical is higher these of other 5 groups. Conclusion Every day 40 mg rana japonica oil configuration low dose compound prescription powder per Rilogiam of body weight perday for big mice its oxidation resistance effect obviously surpasses other dosage of the rana japonica oil compound prescription powder and the squalene capsule.

    Objective To study the feasibility of killing all Anthrax samples by neutron and to work out an effective way in killing Anthrax spore. Method We use MCNP to build a mode and to simulate energy deposition in Anthrax spore per neutron then estimate how much time it takes to kill all Anthrax sample. We also work on the relationship between neutron energy deposition in the anthrax sample and the thickness of the reflector, materials of the reflector, and the area of anthrax sample. Results In this paper, computer simulated data of the largest energy deposition per neutron is 5×10-4MeV/g which appears in situation like:using graphite as reflector, neutron tube for the neutron source, anthrax floor area (10×10) cm2, and 5cm between neutron source and the surface of anthrax. The calculation shows that, for a neutron tube it only takes 20 minutes to kill all the anthrax spores. If using the 1014n/s yield of the neutron tube just ten seconds or longer to reach the lethal dose of anthrax spores. Conclusion To kill the Anthrax spore by neutron is of high efficiency and research value.

    Objective To study the effect of low-dose rate 60Co γ ionizing radiation on rats and provide theory foundation for protection of γ radiation. Methods SD rats were cumulative exposed to 60Co γ-ray. The expose time was 1 hour for every day and dose rate was 0.072 Gy/h. After 7 days, the accumulated dose reached 0.5 Gy and the associated changes in rats were measured. Results After irradiation, the average number of peripheral white blood cells of rats in experimental group(EG) was 3.50×109/L, while that of the rats in control group was 6.2×109/L. The decrease in EG is siginificant (P < 0.05). Other parameters in blood RT including platelet are of No significant difference. The average value of absorbance at OD260 of DNA in marrow of rats in EG was 1.083, while that of the rats in control group was 1.304. The decrease in rats of EG is significant (P < 0.05). The activity of SOD in serum of rats in EG was 373.83 U/ml, while that of the rats in control group was 421.59 U/ml. The decrease in rats of EG is significant (P < 0.05). Compared to the rats in control group, the main organ coefficients of rats in EG showed no significant difference. Conclusion Low-dose γ ionizing radiation have severe impairment effect on hematopoietic system and anti-oxidation system. It has no obviously effect on the organ coefficients of the main organs of rats.

    Objective In order to investigate the effect and the possible mechanism of γ-rays on neurodevelopment of fetal brain tissue as bystander effect organ. Methods pregnant kunming mice were randomly divided into blank control group, 0.5 Gy whole-body exposed group, 0.5 Gy head exposed group, 1.0 Gy whole-body exposed group, 1.0 Gy head exposed group, 2.0 Gy whole-body exposed group and 2.0 Gy head exposed group. The exposed mice were exposed with a vertical single acute dose using 60Co therapy apparatus on the 9 th day of pregnancy, and cesarean operation were performed to gain fetal mice on the 18th day of pregnancy. The number, the size, stillbirth, birth defects and abortion, and get fetal brains from live births were observed. Western-blot assay was used to detect the expression of MeCP2 protein. Results Compared with the blank control group, the rates of stillbirth, birth defects and abortion ascended as the increase of doses; the expression of MeCP2 were upregulated except 0.5 Gy whole-body exposed group, there were no significant differences between groups. Condusion When the pregnant mice were exposed to ionizing radiation in the first trimester, bystander effect in fetal brain tissue was induced, within a certain range,the incidence of deterministic effects and stochastic effects ascended as the increase of doses.

    Objective To analysie the treatment effect of non-small cell lung cancer of three dimensional conformal radiotherapy(3D-CRT) and to study the effect of patient survival related factors. Methods Retrospective analysis was mack for 136 cases of non-small cell lung cancer, all accept 3D-CRT, through the case data collection and long-term follow-up, using the single factor and multiple factor analysis survival time and its influencing factors. Results The recent curative effects of 136 cases of patients with three dimensional conformal radiotherapy:Complete response (CR) 14.7% (20/136), partial response (PR) 60.3(82/136),stable disease (SD) 19.9% (27/136),progression disease (PD) 5.1% (7/136),total effective rate is 75% (102/136). One、two、three、five year survival rate is 79.4%、45.4%、22.1%、12.5%. Side effects:Class 1 radiated esophagitis 35 cases, Class 2 radiated esophagitis 16 cases, Class 3 and above radiated esophagitis 0 case. Class I radiated pneumonia 20 cases, Class 2 radiated pneumonia 9 cases, Class 3 radiated pneumonia 0 case. Single factor analysis shows the influence of gender, age, pathology, phase, dose, and first-phase curative effect to the survival time are of a statistical significane, Multiple factor analysis showed KPS score, phase, dose, first-phase curative effect are the survival time independent factors. Conclusion 3D-CRT for patients with non-small cell lung carcinoma is a safe, effective treatment method, Side effects are relatively low, and the patients survival time is long after radiotherapy.
