    Objective To research on consistency verification of radiation field size based on 2D ion chamber array matrix. Methods We have used a 2D ion chamber array detector (IBA MatriXX) to measure the fields of VARIAN 23EX accelerator. The matrix was lied horizontally on the accelerator couch. The centre of matrix coincided with the accelerator isocen tre,SAD=100cm. The field size was changed from 4.0cm×4.0cm to 16.4cm×16.4cm. Irradiating 6MV-X ray 50 MU per field, the measure data was recorded, and was analysed with the sofware, OmniPRO I' mRT (1.5). Results The difference between the length (X, Y) of light and radiation field displays wave mode cyclicity change with the accretion of light field length. The average distance between the border difference wave crest and trough equals (7.6 ±0.7) mm. The average distance from the trough to the crest in X direction is (3.3 ±0.6) mm, and the average distance from the trough to the crest in Y direction is (3.3±0.5) mm. Conclusion Because the distribution of ion chamber in arrary is discrete and regulative, the difference between the length of light and radiation field displays wave mode cyclicity change with the accretion of light field length. Verifying consistency of radiation field size, the impact of discrete distribution of ion chamber in arrary over measurement result should be considered sufficiently.

    Objective To study changes of NO content,NOS activity and AIF gene in mouse testis induced by low dose radiation.Methods To measure changes of NO content and NOS activity in mouse testis tissue after irradiation with 0~0.2 Gy with enzymic method.At the same time,they were measured after 0~24 h irradiation with 0.075 Gy;We also observe changes of apoptosis inducing factor (AIF)gene in mouse testis tissue after irradiation with 0~0.2 Gy with real-time PCR and Western blot,and AIF mRNA and protein expressions were meausured after 0~24 h irradiation with 0.075 Gy.Results The content of NO and activity of NOS in mouse testis tissue increased with the dose increasing after 12 h irradated with different dose X-rays,it increased with the dose raising and reached the maximum with 0.075 Gy (P<0.05),and kept high level with 0.1 and 0.2 Gy.AIF mRNA increased with dose raising,it reached the peak value when dose was 0.1 Gy;but AIF protein expression reached maximum with 0.075 Gy.The NO content and NOS activity increased with time prolongation after irradiation with 0.075 Gy,NO content reached peak value at 24 h,but NOS activity at 12 h,AIF mRNA and protein increased with time prolongation.mRNA reached maximum at 24 h,but protein at 12 h.Conclusions After irradiation with low dose radiation,NO content,NOS activity and AIF gene expression in mouse testis spermatogenic cells increased and had time and dose effect regularity.

    Objective To investigate the influences for Maitake Polysaccharide on the Immunoregulation and anti-radiating damage.Methods The mice were divided into control group and three experiment groups.The mice of three experiment groups were fed Maitake Polysaccharide at dose of 0.15g/kg、0.30g/kg、0.90g/kg for 30 days.7 indexes were observed to detect the effect of Maitake Polysaccharide on the immunoregulation.The other mice were divided into radiating control group and three experiment groups.The mice of three experiment groups were fed in the same way.Then all animals were radiated by 60Co-γ.The number of white blood cells (WBC)was counted before radiating,on 3 and 14 day after radiating.The DNA content of bone marrow cells were measured on 3 days after radiating.The activities of superoxide dismutase (SOD)in serum were measured on 7 days after radiating.Results Maitake Polysaccharide could increase the amount of hemolytic antibody in serum.The proliferation of ConA-induced mitogenic response for spleen lymphocyte was also increased.Maitake Polysaccharide could enhance the activity of NK cell and the ability of phagocytosis significantly.Compared with the radiating control group,the number of WBC and the DNA content of bone marrow cells,and the activities of SOD in serum increased significantly.Conclusion Maitake Polysaccharide can improve cell-mediated and humoral immune function and non-specific immune function,and protect the mice from radiation injuries.

    Objective To study the change of antioxygen level of those who are continuously exposed to light and chronic radiation.Methods 53 male inhabitants aged from 50-59 years are selected as the object of study,who are born and bred in Yangjiang high natural background radiation area .Additional male inhabitants of similar ages in the town of Hengpi,Enping are chosen as the contrast group.5ml of peripheral blood is separately drawn to determine the activity of superoxide dismutase and the content of malondialdehyde in the serum.Results In the serum of HBRA and CA groups,the activity of SOD is <65.9±38.5> and (50.2±26.2)μ/ml respectively,and the content of MDA is (4.5±2.0)nmol/ml and (6.5±2.6)nmol/ml respectively.Compared with CA group,the activity of HBRA group is high.but its content of MDA is low.These differences are statistically significant (P<0.05).Conclusion Natural high background radiation could have an effect on the antioxidation indexes such as SOD and MDA.

    Objective To investigate the effects of Rad51 depletion on sensitization to γ-radiation in osteosarcoma cell lines.Methods Four human osteosarcoma cells were examined for basal levels of Rad51 by western blotting analysis and RT-PCR.The suppression of Rad51 expression was performed using vector-based shRNA in HOS cells.Sensitivity of HOS cells treated with shRNA vector to γ-radiation was determined with colony formation assay and MTT assay.The apoptosis was determined by Flow cytometry.Results Rad51 showed higher basal levels in osteosarcoma cell lines than those in CCC-ESF-1 both in protein and RNA level.The suppression of Rad51 expression sensitized apoptosis of the cancer cells induced by γ-radiation.Conclusion Rad51 may serve as a promising therapeutic target for osteosarcoma.

    Objective To explore the high-power radar,microwave radiation on rat peripheral blood T-cell subsets.Methods Adult male SD rats were divided into two major groups (for a single experimental study of microwave irradiation and continuous microwave irradiation experimental studies respectively).Each large group according to body weight,was randomly divided into small ones,control group,microwave irradiation group,physical protection groups and drug prevention groups of 10 rats.After irradiation 1d,left ventricular puncture was done and blood sampling was taken to detect changes in blood biochemical indicators and make Data,analysis of variance.Results 1,A single microwave radiation test group:Compared with the control group,① irradiated group ALT was significantly higher (P<0.05);② irradiation group and drug prevention group CK increased significantly (P<0.05);③ irradiation group BUN increased significantly (P<0.05);④ irradiation group LDH increased significantly (P<0.05).2,continuous microwave irradiation test group:Compared with the control group,① irradiated group and drug prevention group CK increased significantly (P<0.05);② irradiation group BUN increased significantly (P<0.05);③ Radiation Groups of LDH was significantly higher (P<0.05).Conclusion Microwave radiation of high-power radar to the body of the heart,kidneys and other organs to a certain extent of the injury,concret expression to make the body bichemical changes.Andolin capsules and anti-electromagnetic radiation fabric have shown good protection effect.

    Objective To establish a feasible method of dose reconstruction with enamel EPR disimetry for medical diagnostic X-ray workers.Methods The usefulness of enamel for dosimetry results from its high content of hydroxyapatite.Carbonate impurities,which are incorporated into or attached to the surface of hydroxyapatite crystals during formation,are converted to CO2- radical through absorption of ionizing radiation.The concentration of radicals increases with absorbed dose.According the concentration of radicals measured by EPR dosimetry method.The enamel dose of medical diagnostic X-ray workers was reconstructed.Results The method of electron paramagnetic resonance (EPR)spectroscopy for tooth enamel is applied to individual dose reconstruction for 14 Chinese medical diagnostic X-ray workers with their entry year before 1985.The enamel doses were ranged from 89 to 870 mGy.Conclusion The method is feasible in dose reconstruction for medical diagnostic X-ray workers.

    Objective To observe radiation Response of cells by WORTMANNIN (WT),which is inhibitor for Phosphatidylinositol-3 Kinase (PI-3K).Methods LP3 cells are prepared with different concentration of WT for 1 hour and receive different dose γ irradiation.To continue the cell for clone culture,and get the production of dose-survival curve.1800 pulsed-field gel electrophoresis is used to detect DNA double-strand breaks after the 20Gyγirradiation.Continue to use the mobility shift assays (Electrophoresis Mobility Shift Assay,EMSA)to observe NF-kB transcription factor of the corresponding changes.Result WT can be found to increase the radiation sensitivity of SP3 cells,the best sensitizer concentration in 20μmol/L or more,obvious sensitizing effect within 6h time;the electrophoresis experiments showed that after irradiation with time,by 50μmol/L WT group DNA the gel is higher than that of the simple exposure group;transcription factor NF-kB binding activity in the 6 hours after exposure experiences a low-rise and then the process of rising with its the peak of the change reaching after about 3 hours after irradiation.Conclusion It suggests the existence of PI-3K-mediated radiation sensitizer pathways.Ionizing radiation may activate NF-kB,which caused some DNA damage repair and other defense mechanisms and cell-related gene activity in order to reduce radiation damage.WT may block this process through the early stages of radiation-sensitizing effect.

    Objective To examine the genetic homozygosity of the IRM-1 mouse bred recently and compare with the IRM-2 and 615 mice to determine IRM-1 mouse genetic characteristics.Methods According to the national stardard GB/T14927.1-2001 and GB/T14927 2-2001 genetic operating instructions,the Akpl etc 25 biochemical markers were detected to observe the genetic homozygosity of the IRM-1 inbred mouse.In addition,the histocompatibility genetic homozygous determination,transplantation experiment between homologous series variant,and hair color test by mating with BALB/c were all carried out.Results All the 25 genetic biochemical markers are homozygote,among which five biochemical markers gene polymorphism sites are different from the IRM-2 mouse,eight biochemical markers gene polymorphism sites are different from the 615 mouse.No dropping graft was found after transplanting 100 days.The coat color genes analysis revealed that hair color of first generation hybrid was wild color,whose gene type was AAbbCCDD.Conclusions The IRM-1 inbred strain mouse is high homozygositous,whose degree has reached international standard.

    Objective To make the public to understand the distribution feature of electromagnetic radiation in nearby regions around the mobile communication base stations,and provide reference for the environmental monitoring department to choose the Monitoring periods and points.Methods Through 24-hour surveys and analysis of electromagnetic radiation in the nearby region around the representative mobile communication base station,the radiant intensity change vs the time and distance in normal operating day.And based on the telephone traffic and Antenna characteristic,the Space-time distribution characteristics of radiant intensity were also studied.Results The result showed that the electromagnetic radiation intensity near the antenna of mobile communication base station had obviously fluctuated character in one day,and the distribution of its power density approached "Μ" type distribution,and it possessed positive relativity with the telephone traffic.Around base stations,the power density showed clear changes with distance,The power density approached "Λ" type distribution in the nearby region.the peak value of power density was related with Antenna characteristic.Conclusions The environmental monitoring department ought to choose the suitable monitoring periods and points according to the telephone traffic and Antenna characteristic,so as to find out the main influence of the basement on the environment of electromagnetic radiation.

    Objective To establish normal reference values of hematology indexes of IRM-2 mice with the different age,and compare with the parental mice and C57BL/6J mice,provide the basic date for the application.Methods WBC、RBC、PLT and BMC were measured by blood routine examination.Results study and identify the normal reference range of the hematology indexes。The result showed that among the male and female IRM-2 mice,there was a significant difference on BMC in the 7weeks and 14weeks old (P<0.01),while there was no significant difference in WBC、RBC and PBC.All Indicators were the top in 14 weeks old,BMC decline in 18 weeks old,and BMC of 56 weeks group becomes lower than that of others.There was significant difference between 18 weeks old group and others group in BMC (P<0.01).The bone marrow nucleated cells and peripheral white blood cells in IRM-2 were greater than those in ICR,615 and C57BL/6J mice (P<0.01)while there was no significant difference in Leukocyte count.Conclusion The hematology indexes of IRM-2 mice were different by gender and age.IRM-2 mice has more powerful hematopoietic,and can be used in biomedical research as a a new inbred strain of mice.

    Objective To investigate the direct effect of X-ray radiation on activity and apop tosis of cultured rat cardiomyocytes in vitro.Methods The study was performed using primary cell cultures of cardiomyocytes isolated from hearts of now-born rats.After being cultured for 4 days in vitro,cardiomyocytes were irradiated with single dose of 10Gy、20Gy、30Gy、40Gy of X-ray respectively.viability of cardiac muscle cells were assessed by MTT colorimetric assay,Myocardial of lactic acid dehydrogenase (LDH)was examined by blood biochemical autoanalyzer.Meanwhile,the Morphological characters of apoptosis were checked with laser-scanning confocal microscope by AO/PI staining and cardiac myocytes apoptosis rates were detected by flow cytometry.Results ①X-ray inhibited the growth of rat cardiac muscle cells in the dose-dependment or time-dependment manner in a certain range of dose.②the Morphological characters of apoptosis induced by X-ray (40Gy)were surveyed by laser-scanning confocal microscope in cultured rat cardiac cells.③X-ray induced apoptosis rate was detected being increased by flow cytometry.Conclusion X-ray has toxic effect,causing the damage of cardiac cells or apoptosis.

    Objective By using laser scanning confocal microscopy (LSCM)to test the dose-effects relationship between ionizing radiation intensity and quantity of the γ-H2AX in vivo and in vitro respectively,and the consistency relationship between the vivo and vitro retrial.Methods To irradiate the peripheral blood from Wister female rats by 137Cs at 7 with different doses (0、0.5、1、2、2.54、4、6、8Gy)extract the lymphocytes from the peripheral blood and detect the dose-effects relationship between irradiation intensity and number of γ-H2AX foci.Results There are good dose-effects relationships between the irradiation and foci number both in vivo and in vitro,which are linear,Yvivo=0.096+0.13X;Yvitro=0.040+0.21X.And there is good consistency (R=0.98)between the γ-H2AX in vivo and in vitro.Conclusions γ-H2AX has the possibility for clinical trial as an indicator,and we can use vitro trials in place of the vivo trails to evaluate the dose people received.

    Objective To investigate the protective effects of fullerenol in ICR mice against 60Coγ-ray.radiation.Methods ICR mice were injected (ip)with fullerenol continuously from 5d before to 7d after whole-body 60Coγ-ray irradiation.The total dose was 5.0Gy and 8.0Gy respectively and the dose rate was 1.0 Gy/min.The change in body weight and index of spleen,index of thymus,peripheral blood cell counts on the 7th day after irradiation were observed.The survival rate and average survival time were monitored during the period of 30 days after irradiation.Result Fullerenol showed significant radioprotective effects on white blood cell and platelet counts,protecting spleen and thymus.Also fullerenol increased the survival rate on the 30th day after irradiation and the average survival time.Conclusion Fullerenol has efficient capacity of radioprotection in irradiated mice.

    Objective To establish models of Hep-2 laryngeal cancer cell line of different oxygen supplying,investigate the impact of hypoxiaon on the apoptosis and cell cycles in Hep-2 human laryngeal cancer cell line induced by different dosage of γ-ray.Methods The cells were diviced into 2 groups:group A (normoxia)and group B (hypoxia).Both of the two group cells are exposured in different dosage of γ-ray which range from 1Gy to 40Gy.Constructinon group (0Gy)was used.Flow cytometry (FCM)was used to measure the distribution of cell cycles and the protein levels of HIF-1α and to detect cell apoptosis.The protein levels of HIF-1α were also determined by immunohistochemistry.Results 1 Gy-5Gy γ-ray caused G0/G1 cell cycle arrest when 10~40Gy γ-ray caused G2/M cell cycle arrest in human Hep-2 cells in group A.All of the different dosage of γ-ray caused G2/M cell cycle arrest in human Hep-2 cell in group B,then without exposure of γ-ray (0 Gy)could cause G0/G1 cell cycle arrest in group B.The curve of apoptosis rate in group A show a parabola,The peak was 41.56±2.25 at the point of 5 Gy.Apoptotic rate was related to the dosage of γ-ray in a dosage dependent manner.The apoptosis rate in group B were significantly lower than that in group A.The dose difference of γ-ray have no impact on the apoptosis rate in group B.Conclutions Different dosage of γ-ray could cause different cell cycles arrest in the condition of normoxia.There was an important relationship between cell cycle arrest and apoptosis.The descend of the apoptosis rate were related to G2/M cell cycle arrest in the condition of hypoxia and the high expression of HIF-1 maybe one of these reasons.
