    Objective To isolate, extract and purify human cardiac myosin (HCM) and human cardiac myosin heavy chain (HCMHC) for it's next study of myocardid image and the antibody against HCMHC imaging reagent.Methods First extract HCM from human heart muscle tissue and prepare HCMHC from the HCM, detecte characteristics including protein concentration, molecular weight, enzymatic activity and free SH groups of the HCM and the HCMHC, prepare hybridoma cell lines of HCMHC, and examine the immunology activity of the HCMHC.Results 24mg HCM were extracted from 127g human cardiac muscle tissue, and 9.1mg HCMHC were prepared.Elecrtrophoresis of SDS-PAGE (gradient) showed that the band of HCMHC located at 200 kD, which was the same of that of literatures, purity of HCMHC is higher than 80%.The specific activity of ATPase of HCM is good.The highest titer of anti-HCMHC serum is about 107.The fusion rate was 80%.Conclusions HCMHC has good immunology activity, for it's next study of the antibody against HCMHC single chain fragment variable (ScFv) radionuclide receptor imaging.

    Objective It is reported that radiation-induced myocardial degeneration in the rat is preceded by changes in capillary structure and function.The aim of the present study is to investigate direct effect of gamma ray radiation on activity and apoptosis of cultured rat cardiomyocytes in vitro.Methods The study was performed using primary cell cultures of cardiomyocytes isolated from hearts of now-born rats.After being cultured for 72h in vitro, cardiomyocytes were irradiated with single dose of 5Gy, 10Gy, 20Gy of gamma ray respectively.At 48h post-irradiation, the concentration of LDH in the supernatant of cell culture was tested using methods introduced by International Federation of clinical chemistry (IFCC), and apoptosis was determined with flow cytometry.The viability of myocytes was determined with crystal violet test and MTT test at 48h and 120h post-irradiation respectively.Results LDH concentration in the supernatant of cell culture of cardiomyocytes were increased significantly with the irradiation dose augment.Flow cytometry confirmed the iuduction of apoptosis in response to different gamma ray doses irradiation at 48h after irradiation.The viable cardiomyocytes irradiated by gamma ray were significantly declined at 120h after irradiation compared to un-irradiated cells, however there were no significant difference between two groups at 48h post-irradiation.Dose-effect relationship was demonstrated between cardiomyocyte apoptosis, viability and irradiation dose in the study.Conclusion The study demonstrates gamma ray radiation can cause direct damage to cultured cardiomyocytes, including inhibiting activity and inducing apoptosis of cardiomyocytes in vitro, which shows dose effect relationship.The mechanism of gamma ray irradiation induced injury to cardiomyocytes shoud be investigated futher.

    Objective To observe the effects of electromagnetic irradiation on the level of GluR2 protein and phosphorrylation in PC12 cells.Methods The cultured PC12 cells were exposed to 90 mW/cm2(average power density) electromagnetic wave for 20 min, The level of GluR2 protein and phosphorylation of PC12 cells were examined by western blot at 0 h, 3 h, 12 h, 24 h after irradiation.Results The level of GluR2 protein of PC12 cells decreased at 0 h and 3 h after irradiation;The level of GluR2 phosphorylation of PC12 cells decreased significantly at 0 h after irradiation.Conclusion 90 mW/cm2(average power density) electromagnetic wave could lower the level of GluR2 protein and phosphorylation in PC12 cells.

    Objective To study the restorative function of compound white peony root oral liquids (cWPROL) in treating rats' experimental acute radiation-induced esophagitis from the aspect of immunodeficiency.Methods 128 Wistar rats were divided into eight groups.The radiation esophagitis was ieduced by 43 Gy 60Co.Then the rats were treated with medicine in different ways.On every experimental point the absolute number and percentage of T lymphocyte subsets were analyzed by flow cytometry.The changes of the leukocyte in number and differential count were detected by haemocyte counting instrument.IgG and C3 in rats' serum were analyzed by immunological turbidimetry.Results Radiation brought on decrease of experimental rats' leukocyte count, the lymphocyte differential count, absolute count and absolute T lymphocyte subsets in the blood and the content of IgG and C3 with radiation esophagitis.cWPROL could increase the lymphocyte percentage compared that in with the radiated group 2.The preventive (high dose) could increase lymphocyte count while Western medicine decreased rats' leukocyte count, lymphocyte percentage and absolute count (P<0.05).The absolute T lymphocyte subsets in rats' peripheral blood increased in the group infused with cWPROL (high dose)(P<0.001).Compared with the radiated group 2 the absolute T lymphocyte subsets decreased significantly (P<0.001), and the content of complement C3 increased in the group infused with cWPROL (high dose)(P<0.05) but decreased in the group infused with Western medicine (P<0.01).Conclusion cWPROL plays the role of restoring the cytoimmunity and immunity damaged by radiation in rats with radiation esophagitis.

    Objective To provide theoretical foundations for KM-HL mice used as experimental mice on leucopenia model.Methods Cyclophosphamide and 137Cs source γ rays irradiated at 7.5Gy once the whole body (dose rate:0.80645Gy/min) were given on KM-HL mice to induce mice leucopenia model, samples were obtained to examine the variation of physiological indexes such as blood, hematopoietic system, immunity, heart and liver etc.Results The research shows, in normal KM-HL mice in the leucopenia experiment, some of the physiological indexes of WBC, DNA, spleen, thymus, heart shows that they are deeply injured (P<0.001), only the Cyclophospamide group's liver is lightly injured, without any statistic difference.Conclusion Either giving cyclophosphamide or 137Cs source γ rays irradiating once all over the body, the physiological indexes such as blood, hematopoietic system, immunity, heart and liver etc of KM-HL mice are descended, compared with the control group (P<0.001), therefore it can be used as experimental mice on leucopenia.

    Objective To establish a rapid method to detect HCMV with ELISA, and to study the infection conditions of radiation workers.Methods using routine ELISA except adding 3% PEG to dilution solution to accelerate the reaction of antibody to antigen, the HCMV-IgG and HCMV-IgM in the serum of radiation workers were detected.Results The total positive rate in 514 ratiation workers in Jinan is 96.69%, of which 94.16% is latent infection, 0.19% is primary infection, 2.34% is relapse infection, and active infection rate is 2.53%.Conclusion Radiation workers have certain risk to suffer from infection of HCMV which should delt with seriousty.

    The influencing factor of measuring radon exhalation rate is studied with the method of the activated charcoal absorption.The sampling time is 24 hours, and water sponge is used to simulate the medium with different water.We use 100g activated charcoal to measure different materials.It is found that when we measure concrete surface, activated charcoal absorbs move than 1g water, educing the measured value by about 1%.When we measure plaster surfaces, activated charcoal absorbs move than 1g water, educing the measured value by about 1.85%.In normal condition, measured value of the radon exhalation rate from plaster surfaces reduces by about 12% and 25% respectively, when we lengthen the sampling time from 24 hours to 48 hours and 72 hours.The anti-diffusion effects are not obvious to the medium which is biggish such in density as concrete.

    Objective To summarize the long-term experience of radioanalysis and solve the current practical problems encountered in the rare earth radioanalysis.Methods Make comparison study by comparing different measurement methods and sample passing analysis between the laboratories.ResultsThe correct selection of standard reference matter is the guarantee of the accuracy of the analysis;γspectrum analysis is the guarantee of rare earth radioanalysis.But as of the non radio-balance samples, other analysis methods should be used simultaneously to get more information from the samples.Conclusion As of some complicated, unknown samples, it is very necessary to make comparison study regularly so as to improve the level of analysis and measurement, and make full use of the national open laboratories.

    Objective To explore the effects of β-carotene from salt algae on mouse thymocytes induced 60Co in vitro.Methods Cell proliferation of the cells was detected by MTT method, protein level, coomassie brilliant blue method, lymphocyte conversion ratio, smear method, and apoptosis, flowcytometry.Results After 4Gy 60Co irradiation, the cell proliferation and the lymphocyte conversion ratio of the thymocytes decreased and apoptosis rate increased significantly.When β-carotene was added into thymocytes before irradiation, cell proliferation and the lymphocyte conversion ratio increased with decreased apoptosis in a dose dependent manner (P<0.05, P<0.01).Conclusion β-carotene has the protective effects on the irradiation damage of mouse thymocytes induced by 60Co in vitro.

    Objective To analyze the effect of fractionated irradiation with variable fraction sizes (VFS) with human mammary tumor cells MCF-7.Methods Ten samples with the same number of MCF-7 cells were irradiated with different fractionated protocols, then using MTT analysis method, the cell survival probability (S) is obtained and compared with each other.Results In nine groups, the S value of the samples irradiated with VFS protocols are lower than that of the sample irradiated with constant fraction sizes (CFS) protocol, also the differences between some of the results are of statistical significance;And for the same variety range, the effect of irradiation with decreasing fraction sizes is better than that of irradiation with increasing fraction sizes, also the differences of S value between two groups are of statistical significance.Conclusion The result of experiment with MCF-7 cells conformes to the former calculation results.Therefore the VFS irradiation protocol, especially the protocol with decreasing fraction sizes can probalely be used in clinics to enhance the treatment effect.

    Objective To investigate the possible threshold between the thorium lung burden and the five hepatic parameters of the miners with long-term inhaled thorium containing ore dusts.Method By using high sensitive negative voltage exhaled thoron progeny collection and measurement system, the thorium lung burdens of each dust exposed miner was obtained.Results Our results showed that whenever the thorium lung burden is below 7.26Bq, the miner's five hepatic parameters would be normal range.Conclusion By our own results and the data from literatures.The possible threshold existing between the thorium lung burden and its effects on five hepatic parameters will be from 7.26Bq~74Bq.
