    Objective To investigate if ionizing radiation could enhance tumor inhibiting effects in mouse.Methods 0.2ml LA795 cell suspension (3.5X107 cells/ml) were subcutaneously injected into flanks of IRM-2 mice, the inoculated mice were devided into 5 groups:control;treated with simple E838 with a dose of 5, 7.5 and 10 mg/kg daily, for 7 days;treated with simple cytoxan with dose at 25 mg/kg (4 times), only irradiated with 1 Gy γ-ray and E838 combined with r-ray group.Results The tumor inhibiting rate for the E838 treated group were 42.73, 70.91 and 64.09% respectively, there were significant difference compared with the control group (P<0.05, The tumor inhibiting rate for the combination group were 63.63, 75 and 68.64 respectively (P<0.05, compared with control group), but there were no significant difference between E838 groups and the combination groups.Conclusion These data suggest that E838 have significant inhibiting effects for inoculated tumor in mouse., the tumor inhibition rate of combination groups seems to be higher than that of E838 treatment groups, but with no significant difference.

    Objective To investigate the effect of 0.4mT power frequency magnetic field on the microfilament (F-actin) assembly of Chinese hamster lung (CHL) cells.Methods F-actin were marked with immunohistochemical method, then observed under a confocal microscope.The content of EGFRs in the preparation of the detergent-insoluble cytoskeleton was measured with Western-blotting.Results The stress fibers of CHL cells decreased after exposure to 0.4mT power frequency magnetic field for 30min, as well as after treatment with epidermal growth factor (EGF) of 50nM.Filopodias appeared at the periphery after exposure to magnetic field as well as treatment with EGF.The EGF receptor mass associated with the detergent-insoluble cytoskeleton increased after exposure to magnetic field as well as treatment with EGF.Conclusion 0.4mT power frequency magnetic field induced assembly of F-actin in CHL cells.The change induced by magnetic field would be related to clustering of EGFR induced by magnetic field and passing the signal down.

    Objective To observe GVHD reaction in mixed BMT.Methods BALB/C mice with acute radiation disease were used as recipient mice models, C57BL/6 mice were taken as donor mice.C57BL/6 mice bone marrow and spleen cells mixed with certain percent of syngeneic spleen cells were underwent mixed BMT to observe their death rate, hematopoietic recovery of WBC lineage, spleen node count, mixed lymphocyte reaction in vivo, chimerism and pathological changes.Results GVHD was relatively strong in non-mixed BMT groups, the group mixed by 3:1 BMT after activation could not alleviate GVHD, whereas the mixed by 1:1 BMT group or by 2:1 group with high percentage of syngeneic spleen cells could lessen the extent of GVHD, as manifested by the quick recovery of peripheral blood WBC count and bone marrow GM-CFU formation, by the alleviation of mixed lymphocyte reaction in vivo as well as by the increase of chimeric rate.Conclusion Mixed BMT could lower the GVHD reaction, which was related to the ratio of syngeneic to allogeneic spleen cells.

    Objective To study the protect effect of Shengmaisan water extract on the small intestine of mice after 60Coγ-whole body irradiation.Methods mice model with irradiative was established by 60Coγ-ray, Shengmaisan water extract was used by clyster once a day of two weeks.14days later, the mice were sacrificed and about 2cm long segment of duodenum, jeSepum and ileum were dissected respectively for crypt survival assay using the technique of crypt counting perunit area.The activity of GSH-Px, SOD and the levels of MDA in the small intestine, the levels of IL-2、IL-4、IL-6 and IL-11 in the serum were measured.Results Shengmaisan water extract (40, 60, 80g·kg-1.bw) can increase crypt survival assay of duodenum, jeSepum and ileum, and the activity of GSH-Px, SOD and decrease the levels of MDA, increase the activity of SOD in model rats and increase the high levels of IL-2、IL-4、IL-6 and IL-11 in the model group in a dose-depended manner.Conclusion Treatment with Shengmaisan water extract can improve the ability to eliminate oxygen free radical and blood interleukin and repair wound.

    Objective To study the effect of diagnostic ultrasound to living rat's embryonic cardiomyocytes on the expression of Bcl-2 and Bax protein and it's apoptosis.Methods To divided the mice which had been pregnant for 14 days into six groups:the 10 min irradiation with 2.5MHz ultrasound the 10 min irradiation with 7MHz ultrasound, the control group of 10 min, the 40 min irradiation with 2.5MHz ultrasound, the 40 min irradiation with 7MHz ultrasound and the control group of 40 min (7MHz probe:Isppa=227W/cm2;Ispta=385mW/cm2;2.5MHz probe:Isppa=94.5W/cm2, Ispta=15.6mW/cm2).Immunohistochemistry test was used to detect Bax and Bcl-2 protein's expression on fetal cardiomyocytes and TUNEL method was used to detect apoptosis.At last, TEM observation was used to observe the morphology of apoptotic cells.Results According to statistics, there had no significant difference in different embryos of the same sample (P> 0.05).Bcl-2 was weakly positive (+) and Bax was weakly positive (+) in 40 min irradiation with 7MHz ultrasound group;Bcl-2 was weakly positive (+) and Bax was negative (-) in 10 min irradiation with 7MHz ultrasound group;Bcl-2 was weakly positive (+) and Bax was negative (-) in 10 min irradiation with 2.5MHz ultrasound group;Bcl-2 was weakly positive (+) and Bax was negative (-) in 40 min irradiation with 2.5MHz ultrasound group;Two control groups were both negative (-) in Bcl-2 and Bax.TUNEL test and TEM demonstrated that 40 min irradiation with 7MHz ultrasound group was more significant than other groups in apoptosis (P<0.05).The rest of the experimental groups had no significant difference than the control groups (P<0.05).Conclusion High intensity diagnostic ultrasound can lead to apoptosis of fetal cardiomyocyte by long irradiation;Bcl-2 and Bax regulated play a regulating role in the process;Small dose of ultrasound can promote the expression of Bcl-2 protein.

    Objective To observe the killing effect of peripheral blood mononuclear cells (PBMCs) irradiated by γ ray on cultured human gastric cancer cell line MKN-28.Methods The experiment were divided into MKN-28 tumor cell control group, PBMCs groups and MKN-28 cells with irradiated or non-irradiated PBMCs co-culture groups.Radidation dosage were from 0.5 to 3 Gy, acridine orange/ethidium bromide (AO/EB) staining were used to observe the kill effect of PBMCs on tumor cells in different period.Results After culture for 144h, the dead cells of several dosage irradiated PBMCs are much more than those of non-irradiated PBMCs group.At 240 hours of culture, the alive PBMCs deareses in number in both irradiated and non-irradiared groups, but decreases in radiated groups are more obvious.After culture for 72 h in the co-cultured groups, the difference is not evident among all radiation dosage groups.After 96~240 h of co-culture, the killing effect of 0.5~2Gy irradiated PBMCs on tumor cells is very strong, especially in 1Gy group, but the killing effect of PBMCs irradiated by 2.5~3Gy on tumor cells were weaker than that of 0.5~2Gy irradiated groups.At 240 hours co-cultured groups irradiated by 2.5~3Gy, tumor cells still survive and proliferate.Conclusion Gamma ray irradiation have killing effect to some PBMCs.The cytocidal effect of PBMCs irradiated by 0.5~2Gy on tumor cells were increased.Chemotaxis and cytocidal effect of tumor cells to postirradiated PBMCs were also found.The killing effect of PBMCs irradiated by 2.5 and 3Gy on tumor cells were restrained.

    Objective To investigate the anti-γ radiation effect of soyisoflavone and soyasaponins.Methods Three batches in the experiment In every batch.clean rank of Kunming male mice were randomly divided into four groups:normal control group, radiated group, soyisoflavone group and soyasaponins group according to their weight.The mice in soyisoflavone group and soyasaponins group were fed with soyisoflavone and soyasaponins for 30 days successively, then the mice were irradiated by 60Coγ-rays in the whole body except those in the normal control group.After radiation, these two groups were fed equally as before.WBC contents, the Granulocytes and the Micronucleated polychromatic erythrocytes in bone-marrow, SOD activities and MDA contents in the blood and liver were measured.Results On the third and the fourteenth day after radiation, the number of WBC in the blood and the number of the Granulocytes in bone-marrow, the activities of SOD in the blood and liver in soyisoflavone group and soyasaponins group were significantly higher than that of radiated group (P<0.05).The number of the Micronucleated polychromatic erythrocytes in bone-marrow and MDA contents in the blood were significantly lower than that of radiated group (P<0.05).Conclusion Soyisoflavone and soyasaponins could prevent the WBC in distal blood and the Granulocytes in bone-marrow decreasing in number after radiation, restrain the production of the Micronucleated polychromatic erythrocytes, improve the antioxidant ability of radiated mice, and release the irradiation injury from γ-rays.

    Objective To investigate the apoptosis of bone marrow leukemia cell in mice after low dose radiation (LDR) at different time and the experimental basis for LDR auxiliary therapy on leukemia.Methods WEHI-3 cells were injected into BALB/c mice through tail veins to make an experimental mice model of myelomonocytic leukemia.60 leukemia mice models were divided half-and half.30 mice models in experimental group were irradiated with LDR of 75mGy at the same time while the others 30 in the control group were not.6 mice models with LDR and 6 mice models without LDR would be killed at the time the 1st day, the 2nd day, the 3rd day, the 5th day and the 10th day after LDR in order to extract bone marrow samples.The apoptosis percentage of leukemia cells in bone marrow was examined.Results The apoptosis percentage of leukemia cells in experimental group was increasing after LDR and went to top on the 2nd day and the 3rd day.The apoptosis percentage of leukemia cells was remarkably different between experimental and control group, all P<0.05.Conclusion LDR could significantly increase the apoptosis percentage of bone marrow leukemia cells in mice.Its mechanism is remarkably different in kill and wound of big dose radiation to tumour cells.It is probably related to of the increase immune exciting response as to promote some cytokine secretion.in leukemia mice.

    Objective To construct recombinant Rb94 gene adenovirus expression vector and to study its effect wirh γ-radiation in inhibiting the growth of breast cancer.Methods Total RNA was extracted from human embryo and reversed transcript to cDNA.Rb94 gene fragment was amplified by PCR and linked to pENTRTM 1A vector.Rb94 gene was recombinated with pAd/CMV/V5-DESTTM by LR recombinant and identified.Combined with γ-radiation, if produces an inhibiting effect on breast cancer cells.Results The recombinant Rb94 gene adenovirus expression vector was constructed successfully.The combined effect is better in inhibiting the malignant cells.Conclusion The recombinant Rb94 gene adenovirus expression vector can be used to investigate the function of Rb94 gene.Combination effect is stronger in inhibiting the growth of human malignant tumor cells.
